A receptor endocitózis inozitol lipid szabályozásának molekuláris alapjai = Molecular basis of inositol lipid regulation of receptor endocytosis
1. A plazmamembrán (PM) foszfatidilinozitol 4,5-biszfoszfát (PtdInsP2) szintjĂ©nek csökkentĂ©sĂ©re kifejlesztett, rapamycin fĂĽggĹ‘ heterodimerizáciĂłs rendszer mĂłdosĂtásával megoldottuk a transz-Golgi foszfatidilinozitol 4-foszfát mennyisĂ©gĂ©nek akut csökkentĂ©sĂ©t Ă©lĹ‘ sejtekben, Ă©s ezzel megnyitottuk az utat e lipid kompartment jelentĹ‘sĂ©gĂ©nek vizsgálata elĹ‘tt. 2. LĂ©trehoztunk egy olyan fehĂ©rje kĂ©szletet, amellyel az 5-15 nm-es tartományban, perces nagyságrendben mestersĂ©ges kapcsolat hozhatĂł lĂ©tre a PM Ă©s az endoplazmás retikulum (ER), a mitokondrium Ă©s az ER, valamint a mitokondrium Ă©s a PM között. Más laborokkal egyĂĽttműködve megállapĂtottuk, hogy a mĂłdszer jĂłl alkalmazhatĂł olyan folyamatok vizsgálatára, amelyben az emlĂtett organellumok egyĂĽttműködve vesznek rĂ©szt (pl. kapacitatĂv Ca2+ beáramlás vagy mitokondriális Ca2+ jel kialakulása). 3. A PM receptorok endocitĂłzisának molekuláris szintű vizsgálatára Ăşj, energiatranszfer mĂ©rĂ©sre Ă©pĂĽlĹ‘ mĂłdszert fejlesztettĂĽnk ki. MegállapĂtottuk, hogy HEK293 sejtekben a Gq fehĂ©rjĂ©t aktiválĂł 1-es tĂpusĂş angiotenzin II Ă©s a 2C tĂpusĂş szerotonin receptor internalizáciĂłját alapvetĹ‘en meghatározza a PM PtdInsP2 mennyisĂ©ge. Kimutattuk, hogy a PtdInsP2 szint csökkentĂ©sĂ©vel kiválthatĂł gátlĂł hatásĂ©rt a klatrin burok lefűzĹ‘dĂ©sĂ©nek elĂ©gtelensĂ©ge tehetĹ‘ felelĹ‘ssĂ©, mĂg a receporok laterális mozgása Ă©s a klatrin burok terĂĽletĂ©n törtĂ©nĹ‘ bekoncentrálĂłdása nem bizonyult PtdInsP2 Ă©rzĂ©kenynek. 4. LĂ©trehoztunk egy sokoldalĂş, nagy Ă©rzĂ©kenysĂ©gű fluoreszcens kĂ©palkotĂł rendszert. | 1. We developed a molecular tool suitable for the acute depletion of the trans-Golgi phosphatidylinositol 4-phosphate pool in live cells by modifying the rapamycin-dependent heterodimerization system. The main advantage of the system is that it allows further investigation of the role of this inositol lipid. 2. We created a protein tool useable for formation of an artificial bridge between organelles such as plasma membrane (PM) and endoplasmic reticulum (ER), mitochondrium and PM, mitochondrium and ER in the range of 5-15 nm in live cells. Collaborating with various laboratories we found the system suitable to investigate processes, in which the mentioned organelles worked in coorporation (e.g. store operated Ca2+ influx, mitochondrial Ca2+ signal). 3. Based on energy transfer measurements we developed a new method to follow the endocytosis of PM receptors at molecular level. We found that internalization of the Gq coupled type 1 angiotensin II recepors and type 2C serotonin receptors was highly determined by the PM PtdInsP2 content in HEK293 cells. We showed that the failure of the fission of clathrin coated pits were responsible for the inhibitory effect evoked by the decrease of PM PtdInsP2 concentration, while the lateral movement of the receptors, as well as their concentration in the clathrin coated membrane were not affected by the PtdInsP2 depletion. 4. We set up a versatile, highly sensitive digital image system suitable for single cell fluorescent measurements
A prolaktin-releasing peptid szerepének vizsgálata a központi idegrendszerben = Role of prolactin-releasing peptide in the central nervous system
A prolaktin-releasing peptid (PrRP) számos, központi idegrendszeri funkciĂłját tanulmányoztuk. A tejelválasztásban a PrRP-t nem találtuk fontosnak. KrĂłnikus, de nem akut stressz után a PrRP expressziĂł megnĹ‘tt, az adaptáciĂł során megváltozott szabályozási mechanizmusra utalva. A PrRP expressziĂłban Ă©s stresszre adott válaszreakciĂłjában nem-Ă©s genotĂpus-fĂĽggĹ‘ változásokat találtunk. NĹ‘stĂ©ny patkányok agytörzsi PrRP-termelĹ‘ sejtjeiben ösztrogĂ©n receptort (alfa tĂpust, bĂ©tát nem) mutattunk ki, mely magyarázhatja a hĂmekĂ©nĂ©l magasabb PrRP expressziĂłt Ă©s enyhĂ©bb PrRP stressz-választ. Vazopresszin-hiányos Brattleboro patkányokban magasabb agytörzsi PrRP mRNS expressziĂłt Ă©szleltĂĽnk, ami a kontrolloktĂłl eltĂ©rĹ‘en akut stresszre az A2 katekolaminerg sejtcsoportban fokozĂłdott. Agytörzsi, organotipikus szövetkultĂşrán vĂ©gzett in vitro kĂsĂ©rletekben ugyanezen a terĂĽleten a vazopresszin agonista a PrRP mRNS expressziĂłt gátolta. Számos PrRP-pozitĂv idegvĂ©gzĹ‘dĂ©st találtunk a dorsolateralis hipotalamuszban, szinaptikus kontaktusokat lĂ©tesĂtve melanin-koncentrálĂł hormon-(MCH) Ă©s orexin A-termelĹ‘ idegsejtekkel. Az MCH sejtek legtöbbje Fos immunpozitivvá vált 72h REM alvásmegvonást követĹ‘ 3h pihenĂ©s hatására. Az orexin A pozitĂv sejtek nagy rĂ©sze aktiválĂłdott 12h hideg-stresszt követĹ‘ 3h pihenĂ©s után. Az aktivált MCH-Ă©s orexin A-pozitĂv neuronok közĂĽl sok állt közeli kontaktusban PrRP-pozitĂv vĂ©gzĹ‘dĂ©sekkel. Munkánk során sokakat Ă©rintĹ‘ metodikai problĂ©mákat is megoldottunk, eredmĂ©nyeit publikáltuk. | Several aspects of prolactin-releasing peptide?s (PrRP) functions in the CNS were studied. Role of PrRP was not confirmed in lactation. PrRP expression elevated after chronic but not after acute stress indicating a switch in the regulatory mechanisms during the adaptation. Gender- and genotype-specific differences were found in the PrRP expression and in the PrRP response to stress. Estrogen alpha, but not beta receptor was demonstrated in the medullary PrRP neurons of females that may explain the females? higher PrRP expression and less sensitive PrRP reaction to chronic stress. Vasopressin-deficient Brattleboro rats showed higher medullary PrRP mRNA expression then the controls, and unlike controls acute stress evoked PrRP mRNA expression in the A2 cell group. In vitro experiments on brainstem organotypic slice cultures showed inhibitory effect of vasopressin agonist on PrRP mRNA expression in the same area. In the dorsolateral hypothalamus PrRP-positive terminals established synaptic contacts with melanin-concentrating hormone (MCH)- and orexin A-like neurons. Most of the MCH neurons were Fos-positive after 72h paradoxical sleep deprivation followed by 3h rebound. A great percent of the orexin A neurons were activated by 12h cold followed by 3h rebound. Many of the activated MCH and orexin A positive neurons formed close contacts with PrRP-positive terminals. A Renox szuperoxid-termelő enzim vizsgálata emlős sejtekben = Characterization of the Renox superoxide-producing enzyme in mammalian cells
1. KifejlesztettĂĽnk egy Nox4 (Renox)-deficiens egĂ©rmodellt. Az eddigi vizsgálataink szerint a Nox4 jelenlĂ©te nem nĂ©lkĂĽlözhetetlen az Ă©lethez Ă©s az állatok konstitutĂv vĂ©rkĂ©pzĂ©se is normálisnak tűnik. 2. Kimutattuk, hogy az eddig ismeretlen funkciĂłjĂş p50RhoGAP fehĂ©rje fontos szerepet játszhaz a Rho Ă©s a Rab G-fehĂ©rjĂ©k jelátviteli Ăştvonalainak összekapcsolásában. 3. Dr Thomas Leto laboratĂłriumával egyĂĽttműködĂ©sben megállapĂtottuk, hogy a Rac1 kismĂłlsĂşlyĂş GTP-kötĹ‘ fehĂ©rje fontos szerepet játszik a Nox1-et Ă©s Nox3-at tartalmazĂł NADPH oxidázok szabályozásában. 4. A Nox4 Ă©lettani funkciĂłját kutatva sikerĂĽlt azonosĂtanunk egy olyan Ăşj emberi peroxidázt amely fontos szerepet játszhat a reaktĂv oxigĂ©nszármazĂ©kok hatásának közvetĂtĂ©sĂ©ben. | 1. We have developed a Nox4 (Renox)-deficient mouse model. Preliminary analysis of this model indicates that the function of Nox4 is not essential for life and Nox4 has no role in the regulation of constitutive haematopoiesis. 2. We have shown that the p50 Rho-GAP protein provides a link between Rho- and Rab GTPases. 3. In collaboration with Dr Thomas Leto we have shown that the small GTPas Rac1 has an important role in the regulation of Nox1- and Nox3-based NADPH oxidases. 4. While searching for the physiological function of Nox4 we have identified a novel human peroxidase which might have an important role in the mediation of the effects of reactive oxygen species

A kutatási eredmĂ©nyeket „A világ nyelvi kĂ©pe. (Tinta KönykiadĂł, Budapest, 2008. pp. 356); these results can be summarized as follows: 1. we outlined the theoretical framework of the linguistic image of the world as of a cognitive conceptual structure, with respect to the specific character of language and culture; 2. we showed the role of language in the creation of the linguistic image of the world; 3. in the Hungarian language we reconstructed the linguistic image of the concepts marked by the following words and expressions e.g.”mother”, „father”, „family”, „head”, „hand”, „earth”, „homeland”, „death”, „positive feelings e.g. joy, serenity, gaiety, happiness, ecstasy”, „negative feelings e.g. horror, dread, shudder, shiver, fright, panic, excitement, anxiety, „national stereotypes”, „linguistic communication model” etc.; 4. we analysed the metainformational operators being the components of the communication fragments as of the basic unities of usage (e.g. the metainformational markers of meaning in texts; conjunctions; metainformational sentences in the onset of texts; „I see, yes, what”, metainformational verbs etc.); 5. we also touched upon the question that the reconstruction of the linguistic image of the world amounts to a new task for semantic research and puts the nature, interpretation and functioning of human language in different new light; 6. we also touched some aspects of the Polish-Hungarian contrastive semantics
Quantifying lipid changes in various membrane compartments using lipid binding protein domains
One of the largest challenges in cell biology is to map the lipid composition of the membranes of various organelles and define the exact location of processes that control the synthesis and distribution of lipids between cellular compartments. The critical role of phosphoinositides, low-abundant lipids with rapid metabolism and exceptional regulatory importance in the control of almost all aspects of cellular functions created the need for tools to visualize their localizations and dynamics at the single cell level. However, there is also an increasing need for methods to determine the cellular distribution of other lipids regulatory or structural, such as diacylglycerol, phosphatidic acid, or other phospholipids and cholesterol. This review will summarize recent advances in this research field focusing on the means by which changes can be described in more quantitative terms. © 2016
